Variance component model to account for sample structure in genome-wide association studies.

Text

Under certain conditions, one can expect the variance of the test statistics to be inflated by a constant across the genome7,8. A formal model of hidden relatedness based on the coalescent theory1 also suggests a constant inflation across the genome when the sample structure is entirely due to hidden relatedness7. However, for a more complex genealogical relationship among individuals, it is not clear how the inflation of test statistics will behave.
