interneurons revealed that NKX2-1 expression was negligible in hCOs-derived interneurons (Figure 5F). Some neurodevelopmental genes (e.g. MAP2 and ALCAM) were enriched in hMGEOs-derived interneurons, whereas hCOs-derived interneurons displayed higher cortical developmental genes (e.g. ZIC1, PTN and MEIS2) (Inoue et al., 2007; Manuel et al., 2015) (Figure 5G), indicating the differential identities among hMGEOs- and hCOs-derived interneurons. Three neuronal clusters show no enrichment of markers for interneuron or cortical neuron, and thus were annotated as non-committed neurons (Neuron1-3). These clusters were from both hMGEOs and hCOs, and decreased with further organoid development (Figure 5H).
