0.01 (Fig 3I) and the number of VGAT synapses per dendritic soma (MAP2) was 103±18 (Fig 3J). Glutamatergic neurons in the cultures were identified by co-localization of presynaptic vesicular glutamate transporter 1 (VGLUT1) with synaptophysin1 puncta, and by presence of glutamatergic post-synaptic marker HOMER1 puncta (Fig 3K–3M). The fraction of all VGLUT1 puncta to synaptophysin1 puncta on MAP2-positive dendrites was 0.61 ± 0.05 (Fig 3N) and the number of VGLUT1 synapses per dendritic soma was 255±89 (Fig 3O). These data indicate that the hPSC-derived neurons form a network with both GABAergic and glutamatergic synapses.
