Three-dimensional analysis of dendritic morphology was performed using Neurolucida (MBF Biosciences Inc.) with the experimenter blind to treatment. 3–5 neurons per animal per region were traced using Neurolucida at 100x creating a three dimensional reconstruction of each neuron including the full dendritic arbor with spines (Figure 1). Layer II/III pyramidal neurons were identified by triangular soma, and apical dendrite. Medium spiny neurons were identified by soma size, many dendritic extensions and spines as described previously (Robinson and Kolb 1997). Fully reconstructed neuronal tracings (see Figure 1) were analyzed in the companion software, Neurolucida Explorer.
