In summary, in this study, by combining functional analysis, immunocytochemistry, and computer modeling, we uncover the molecular basis and mechanism underlying the activation of STOCs by Ca2+ sparks. Because uncoupling of Ca2+ sparks and STOCs contributes to disorders such as hypertension, overactive bladder, and retinopathy, the results and approaches presented in this study could be useful in understanding these disorders and local Ca2+ signaling in general.
