only slightly attenuated (p=4·66 × 10−11). The novel signal at TET2 (rs2047409) also showed strongest association in never smokers (p=1·31 × 10−8). rs2047409 is separated from the previously reported association of rs10516526 with FEV1 (GSTCD)5,7 by a recombination hotspot and is statistically independent (rs105165267 included as a covariate; rs2047409, p=9·8 × 10−9). TET2 encodes tet methylcytosine dioxygenase 2, which has a role in myelopoiesis, and SNPs in TET2 have shown association with height.36 TET2 was differentially expressed during fetal lung development (appendix pp 58–59).
