In order to model the effects of ethanol in humans, we used an iPS cellular model derived from individuals with no known alcohol dependence. We chose to treat the cells for either 24 hours (24hr, acute) or 7 days (7d, prolonged) with a dose of ethanol (70mM) that is consistent with the blood alcohol content following an episode of binge drinking or chronic alcohol consumption (Centers for Disease Control and Prevention (CDC), 2004) [19] and that triggers cortical neurodegeneration in fetal alcohol syndrome [20].
