We wished to explore whether allele-specific correlation analyses can identify gene sets and pathways associated with risk genotypes that are potentially informative as to underlying pathogenic mechanisms. A notable treatment-specific cis-eQTL was observed at rs3859192 to GSDMA, most significantly after 2-hour LPS (P = 2.6 × 10−52) (Fig. 6, E and F). This SNP is associated with white cell count (49) and asthma susceptibility (50), and is a known eQTL in lung resections (51). The CEU genotype frequencies of this SNP are 0.257:0.496:0.248 (CC/CT/TT), making it an ideal candidate for exploring underlying allele-specific regulation.
