Many studies have examined the A118G SNP and response to NTX in individuals with AUD who are of European ancestry.29–35 For example, Oslin et al. conducted a study examining the association of A118G polymorphism and response to NTX; they found that A/G and G/G carriers had lower rates of relapse and increased time to heavy drinking over the 12-week treatment period compared to A/A carriers, suggesting that this polymorphism can be used to identify individuals who are more likely to benefit from treatment with NTX.29 Similarly, data from the Combined Pharmacotherapies and Behavioral Interventions for Alcohol Dependence (COMBINE) study from individuals of European ancestry who provided DNA samples were used to determine the ability of this polymorphism to predict NTX treatment response.32 Subjects with an Asp40 allele experienced more days abstinent and fewer heavy drinking days with medical management (MM) and NTX compared with medical management and placebo; there was no difference observed between treatment groups in individuals with the Asn40/Asn40 genotype.
