This study investigates the genetic association between the five personality traits N, E, O, A and C and MDD, as well as BD. On the basis of the results of a GWA meta-analysis of N, E, O, A and C in >13 000 subjects,25 individual polygenic scores were calculated and tested for their effect on case–control status in 2 combined target MDD samples and in 2 combined target BD samples totaling 8921 and 6329 subjects, respectively. We first asked whether the genetic association between N and MDD as found in the twin studies is confirmed using polygenic score analysis and to what extent the other personality traits are genetically associated with MDD. Second, we investigated the genetic relationship between personality and BD. Finally, we asked what the differences are between the genetically mediated personality profiles underlying BD and MDD.
