First, we confirmed that the expression levels of DGCR8 in patient-derived neurospheres were reduced compared with those from controls (P=0.0423) by real-time quantitative RT-PCR (Figure 3a), although there was not a large difference of DGCR8 expression between control and patient fibroblasts or hiPSCs (Figure 3a). Next, we performed microarray-based miRNA analysis and measured the expression levels of 2042 human mature miRNAs using the miRBase Rel. 19.0 platform in control and patient-derived neurospheres. We found that 19 miRNAs satisfied our differential expression criteria of an absolute fold change (FC) <0.83 or >1.2 and P<0.05 (Figure 3b and Supplementary Figures 3a and 3b).
