difference between the MTF1-3′ UTR-mutant-type (Mut) + LINC00665 and control groups. After treatment with actinomycin D, quantitative real-time PCR was used to compare the half-life of MTF1 mRNA in the two groups, showing a significantly shortened half-life in the LINC00665+ group and a prolonged half-life in the STAU1− group (Figures 3E and 3F). Considering the potential involvement of UPF1 in the SMD pathway, we also measured the half-life along with the mRNA and protein expression levels of MTF1 in glioma cells with UPF1 knockdown, demonstrating the expected results (Figures S3A–S3C).
