Expanding the study of interactions to the whole genome, Kam-Thong et al. (2012) presented a whole genome analysis of epistasis (SNP-by-SNP) on several traits, including hippocampal volume. Pandey et al. (2012) reported a pathway analysis, which includes information from the network of gene–gene interactions as well as main effects when prioritizing genes for pathway analysis. Clearly no genetic variant acts alone; however, multiple comparisons increase exponentially with this approach implying difficulty obtaining enough power.
