UGT2B10 plays a key role in nicotine and cotinine glucuronidation, converting nicotine to nicotine-glucuronide (a minor nicotine metabolic pathway, accounting for 3–5% nicotine clearance111213), and cotinine to cotinine-glucuronide (accounting for 12–17% of cotinine clearance11). The minor allele at rs144647471 (i.e., that associated with higher cotinine levels in our sample) is associated with a reduction in UGT2B10 function for both nicotine and cotinine glucuronidation1415.
