Overall, we demonstrated that all four comorbid case groups (depression with anxiety, bipolar disorder, schizophrenia or SUD) have increased polygenic burdens of common risk variants for several psychiatric phenotypes compared to the non-comorbid depression case groups (overall mvPRS p-values: PDEP-ANX=1.2 x 10−21, PDEP-BP=1.5 x 10−15, PDEP-SZ=1.7 x 10−15, PDEP-SUD=6.4 x 10−97), revealing an overall PRS pattern that distinguishes the comorbid case groups from their non-comorbid counterparts. We also note that all three comorbid subgroups showed an increased load of DEP-PRS compared to the depression-only case groups.
