Stbl3 cells; incubate the mixture on ice for 10 min, heat-shock it at 42 °C for 30 s, return it immediately to ice for 2 min, add 100 μl of SOC medium and plate it onto LB plates containing 100 μg ml–1 ampicillin. Typically, there are no colonies on the negative control plates (ligation of vector only, with no Sanger amplicon insert), and tens to hundreds of colonies on the experimental plates. Pick a minimum of 48 clones to inoculate in 3 ml of LB-ampicillin culture.
