Prior to beginning, Matrigel-coated plates were prepared as described in Section 2.1.1. NPC medium, DMEM and Accutase (Sigma #A6964) were pre-warmed to room temperature, and the Matrigel–DMEM coating mixture replaced with 2 ml of NPC medium.
