Given the role that the amygdala plays in fear conditioning, Xu and collaborators evaluated GAT1508 in rats using a Pavlovian conditioned fear paradigm, a preclinical model of post-traumatic stress disorder [41]. GAT1508 (10 and 30 mg/kg i.p.) facilitated fear extinction, without affecting cognitive or motor functions (Figure 3) [40]. In short, this new small molecule shows increased selectivity for GIRK1/GIRK2 channels, overcomes the undesired cardiac effects, and has great promise as a potential therapeutic for the treatment of post-traumatic stress disorders.
