selection activity of the retromer CSC with tubule formation (Purushothaman et al., 2017). Yeast do not have an obvious TBC1D5 homologue, however, and the yeast retromer complex is a much tighter association of the SNX-BAR dimer with the retromer CSC than has been observed in mammalian cells (Harbour and Seaman, 2011; Seaman et al., 1998). Therefore, it seems likely that the increased association of Rab7a with the retromer CSC in mammalian cells after loss of TBC1D5 will not dramatically alter the association of the retromer CSC with the SNX-BAR dimer in mammals as this is already a very transient association (Swarbrick et al., 2011).
