Four CtBP2 promoter fragments (−2000/−1 bp, −1307/−1 bp, −675/−1 bp and −1350/−652 bp from the transcription start site) were isolated by PCR amplification and ligated into the luciferase reporter vector pGL3-Basic from Promega (Fitchburg, WI, USA). Huh7 cells were plated on 24-well plates one day prior to the transfection. Huh7 cells were co-transfected with luciferase promoter reporter vectors and GLI1 expressing plasmids. After 48 hours, luciferase activity was examined using the Dual-Luciferase Reporter Assay System (Promega) according to the manufacturer's protocol.
