After first establishing the pluripotency of all three iPSCs generated from each of the Family-811 quartet members through multiple methods, and demonstrating a lack of an overall differentiation deficit into multiple germ layers, our results point to specific defects in this family in the development, establishment or maintenance of NPCs, potentially due to the dysregulation of PAX6, an essential regulator of neurogenesis and corticogenesis.
