500 ng of total RNA for each sample was amplified and purified using the Illumina TotalPrep-96 RNA Amplification kit (Life Technologies, UK), according to the manufacturer’s instructions. Biotin-Labelled cRNA was then normalized to a concentration of 150 ng/ul and 750 ng was hybridised to Illumina Human-12 v4 BeadChips (Illumina, CA, USA) for 16 hours (overnight) at 58°C. Following hybridisation, BeadChips were washed and stained with streptavidin-Cy3 (GE Healthcare, UK). BeadChips were then scanned using the BeadArray reader and image data was then processed using GenomeStudio software (Illumina, CA, USA).
