Text

We have identified (from the analyses above) 45 significant causal associations between health risk factors and diseases (Fig. 2). As the risk factors are not independent, we further sought to estimate the effect of a risk factor on a disease adjusting for other risk factors. To do this, we first investigated the causal associations among the risk factors. We detected 19 significant associations by the GSMR analysis among the 7 risk factors at a FWER of 0.05 (PGSMR < 1.2 × 10−3) (Supplementary Fig. 13). For example, BMI had a significant negative effect on HDL-c (\documentclass[12pt]{minimal} \usepackage{amsmath} \usepackage{wasysym} \usepackage{amsfonts} \usepackage{amssymb} \usepackage{amsbsy} \usepackage{mathrsfs} \usepackage{upgreek} \setlength{\oddsidemargin}{-69pt} \begin{document}$$\hat b_{xy} = - 0.29$$\end{document}b^xy=-0.29), and positive effects on TG (\documentclass[12pt]{minimal} \usepackage{amsmath} \usepackage{wasysym} \usepackage{amsfonts} \usepackage{amssymb} \usepackage{amsbsy} \usepackage{mathrsfs} \usepackage{upgreek} \setlength{\oddsidemargin}{-69pt} \begin{document}$$\hat b_{xy} = 0.28$$\end{document}b^xy=0.28) and DBP (\documentclass[12pt]{minimal} \usepackage{amsmath} \usepackage{wasysym} \usepackage{amsfonts} \usepackage{amssymb} \usepackage{amsbsy} \usepackage{mathrsfs} \usepackage{upgreek} \setlength{\oddsidemargin}{-69pt} \begin{document}$$\hat b_{xy} = 0.15$$\end{document}b^xy=0.15).
