In Figure 2A, the three lines depict statistical power for three different possible cG×E effect sizes. As a point of reference, the effect size designations in Figure 2A reflect what would be considered very large (r2=0.10), large (r2=0.01), and moderate (r2=0.001) for genetic main effects in large GWA studies, which provide the most reliable information about the true effect sizes of genetic main effects (31). We used these effect sizes to provide points of reference, although it is possible that G×E effects tend to be larger or smaller than genetic main effects.
