Multiple lines of investigation have examined neuronal development in brain specific organoids to date, mostly focusing on cortical development. Midbrain- and hypothalamus- specific organoids were also generated (Jo et al., 2016; Qian et al., 2016), but methods to derive organoids resembling other brain areas are less developed, with the exception of two very recent studies reporting on ventrally specified organoids, which we discuss further below (Bagley et al, 2017; Birey et al, 2017). Here, we describe strategies to generate human brain organoids mimicking the MGE or cortex domain. Notably, we describe a robust method for generating human MGE organoids to study human interneurogenesis and migration in 3-D cultures.
