Twelve-week-old iPSC derived neural cultures contained a mixture of MAP2-postive neurons and GFAP-positive astrocytes (Figure 1A), with no overall morphological differences observed between neural cultures derived from CTLs and ADs. About half of the total cells in these cultures expressed TBR1, a transcription factor marking excitatory forebrain-type glutamate neurons. The percentage of TBR1+ cells did not differ between 6 CTL and 6 AD neural cell lines (p = 0.94) (Figure 1B–C).
