Arc puncta were quantified using ImageJ software by first thresholding 8-bit images to 50-70 minimum pixel values to reduce background signal. Particles were counted with 0.2-2 μm2 area and 0.5-1 circularity. These parameters were determined by comparing Arc images with sense control images.
