The asymmetry in the confidence interval suggests that a normal approximation for \documentclass[12pt]{minimal} \usepackage{amsmath} \usepackage{wasysym} \usepackage{amsfonts} \usepackage{amssymb} \usepackage{amsbsy} \usepackage{mathrsfs} \usepackage{upgreek} \setlength{\oddsidemargin}{-69pt} \begin{document}$$\hat \beta _{GY}$$\end{document}β^GY would be inappropriate. We therefore generated an empirical distribution of \documentclass[12pt]{minimal} \usepackage{amsmath} \usepackage{wasysym} \usepackage{amsfonts} \usepackage{amssymb} \usepackage{amsbsy} \usepackage{mathrsfs} \usepackage{upgreek} \setlength{\oddsidemargin}{-69pt} \begin{document}$$\hat \beta _{GY}$$\end{document}β^GY to assess its significance (Supplementary Note 1). Of 10,000 simulations of \documentclass[12pt]{minimal} \usepackage{amsmath} \usepackage{wasysym} \usepackage{amsfonts} \usepackage{amssymb} \usepackage{amsbsy} \usepackage{mathrsfs} \usepackage{upgreek} \setlength{\oddsidemargin}{-69pt} \begin{document}$$\hat \beta _{GY}$$\end{document}β^GY, none were less than zero, suggesting that rs35705950 has a positive log hazard ratio with P-value of order less than 10–4. The empirical 95% confidence interval for the log hazard ratio was 11.58–126.56, suggesting an extremely strong effect on survival. However, in view of the substantial uncertainty in \documentclass[12pt]{minimal} \usepackage{amsmath} \usepackage{wasysym} \usepackage{amsfonts} \usepackage{amssymb} \usepackage{amsbsy} \usepackage{mathrsfs} \usepackage{upgreek} \setlength{\oddsidemargin}{-69pt} \begin{document}$$\hat \beta _{GY}$$\end{document}β^GY, we refrain from drawing a strong conclusion beyond the direction of effect.
