Hippocampal neurons were cultured on coverslips from E18 rat hippocampus as previously described (Roche and Huganir, 1995). The neurons were transfected at 7 days in vitro (DIV). ~20 days after transfection, neurons were incubated with GluA1 antibody and then fixed. After blocking the neurons were incubated with the Alexa Fluor 555–conjugated secondary antibody. The neurons were mounted and imaged under a Zeiss LSM 710 confocal microscope.
