higher from week 3 onwards compared to monocultures (Fig. 3b). All measured parameters reached a plateau 4 to 5 weeks after final plating and remained high at least up to week 8 (Fig. 3c). The individual contributions of DAPT treatment and astrocyte co-culture were questioned further. Co-culture with astrocytes (without DAPT) did not improve any of the parameters, while DAPT treatment (without astrocytes) only increased the amount of active neurons. Therefore, the optimal condition is co-culturing with DAPT.
