Genes with the highest degree of connectivity within a module are termed hub genes and are expected to be functionally important within the module. M8 hub genes included transcription factors TBR1, FEZF2, FOXG1, SATB2, NEUROD6, and EMX1 (Fig. 4b), which have been functionally implicated in NCX and HIP development4,30-38. Furthermore, FOXG1 variants have been linked to Rett syndrome and intellectual disability34. Conversely, M15 GO categories included ionic channels (P=8.0×10-8) and neuroactive ligand-receptor interaction (P=4.0×10-14). Sequence variants in hub genes (Fig. 4c) have been linked to major depression (GDA)39 and to schizophrenia and affective disorders (NRGN and RGS4)7,40.
