In contrast, decitabine withdrawal led to re-establishment of global DNA methylation (5mC) at D8 as detected by IF (Figure 4F). Furthermore, DNA dot blot assay showed DNA methylation reprogramming upon decitabine withdrawal, with elevated 5mC levels (1.5-fold, P <0.05), and relatively unchanged 5hmC levels in decitabine-treated NSC compared to controls (Figure 4G-H). Although globally altered 5mC levels were expected following decitabine treatment in agreement with previous studies [54,55], the observed effect of decitabine to slightly increase 5hmC levels was novel and might be biologically important.
