The full set of 2.8 million (2501×1100) Pearson correlations is then estimated and corrected for multiple comparisons. Bonferroni correction, which is likely to be somewhat conservative in such situations, due to non-independence across variables tested, resulted in pcorrected<0.05 being equivalent to requiring puncorrected<1.8×10-8. An alternative popular approach for multiple comparison correction is false discovery rate (FDR);82 we use the more conservative FDR option (making no assumption of variable dependencies83), resulting here in requiring puncorrected<3.8×10-5. These two threshold levels are shown with dotted lines in all Manhattan plots in the main figures.
