Salivary samples were obtained on each session (both deprived and non-deprived), 5 minutes after spray administration prior to the habituation block (placebo only) and the test block (placebo/nicotine). The samples were used to provide estimates of nicotine exposure between conditions and as a function of smoking deprivation. Assessments of cotinine in saliva samples were obtained on the first non-deprived session to provide an overall measure of tobacco exposure for characterizing the sample. The samples were analyzed for nicotine and cotinine using the High Performance Liquid Chromatography (HPLC) method of Hariharan & VanNoord (Hariharan & VanNoord, 1991). The assay provides sensitivity to <1ng/ml of nicotine and cotinine.
