We used the FUMA and MAGMA pipeline to conduct gene implication and signal pathway analysis of the newly-identified locus associated with OUD. Through position mapping with a 20-kb distance window, four protein coding genes were mapped for the newly-identified SNP on chromosome 4, including DEFB131, SLC2A9, RP11–1396O13.13 and ZNF518B (Supplemental Table S1). Among these, DEFB131 has been reported to be associated with immune function and SLC2A9 plays roles in gout and Alzheimer’s disease(Kim et al., 2015; Ruiz et al., 2018). In the exposed-control analysis, no protein coding genes were mapped close to the locus on chromosome 16.
