As another part of validation, we checked our top SNPs (Table 3) in the summary statistics of Polimanti’s 2020 study(Polimanti et al., 2020). Among 5 SNPs we checked, rs10014685, rs114237113, rs2001392 and rs12931235 were not identified in their SNP list, which could be caused by different pre and post imputation quality control pipelines. rs12596025 showed a non-significant signal (p-value = 0.1698) in their study cohort of opioid dependence cases vs. opioid-unexposed controls of European-ancestry. We also calculated the genetic correlation between our study and the cohort of European ancestry from Polimanti’s results (opioid dependence cases vs. opioid-unexposed controls), but did not observe a significant correlation (rG=1.00, se=0.96, p=0.24), while large SE's indicate that this analysis is underpowered to draw reliable conclusions.
