Chromatin was obtained from CD4+CD25− T cells, CD8+ T cells, CD19+ B cells, and CD56+ NK cells of two healthy male donors each. Chromatin immunoprecipitation (ChIP) for H3K4me1 and H3K27ac and library construction were done essentially as described elsewhere52. Sequence tags were mapped to the current human reference sequence (GRCh37/hg19) using Bowtie50 and only uniquely mapped tags were used for downstream analyses. H3K4me1 and H3K27ac ChIP-seq data for CD14+ monocytes was generated elsewhere52. Complementary DNase hypersensitivity sequencing data was obtained from the Epigenetics Roadmap project (www.roadmapepigenomics.org) and mapped as above.
