A 2 cm2 area of skin was shaved in the middle of the back of 6 to 10-week-old mice and a solution of 5% 1-chloro-2,4-dinitrobenzene (DNCB, Sigma) dissolved in acetone/olive oil was applied. Five days after this sensitisation treatment the mice were rechallenged by applying 0.5% DNCB on each side of both ears. The skin response was assessed 24 h after rechallenging by measuring the interfollicular thickness of the epidermis, hematoxylin and eosin (H&E) staining and histology, as well as immunostaining with specific antibodies.
