The wildtype BAF53b was V5 tagged and cloned into the pAAV-IRES-hrGFP Vector (Agilent Technologies) under the CMV promoter. Adeno-associated virus (AAV) serotype 2/1 for AAV-Baf53b was purchased from Penn Vector Core; University of Pennsylvania. AAV-hrGFP serotype 2/1 was generated and viral titer was determined by qPCR with primers specific to hrGFP57. A titer of 1010-1013 genome copies/ul was used for all injections.
