TNFR1 transcript levels were measured by real-time quantitative PCR (qRT-PCR) with an ABI Prism 7900HT Sequence Detection System and analyzed using SDS v.2.4 software (Applied Biosystems). GAPDH house-keeping gene transcripts and transcripts containing exons 6 and 7 were detected using Taqman® Gene Expression Assays Hs02758991_g1 and Hs00533565_g1, respectively (Applied Biosystems). A custom primer/probe set spanning the exon 5-7 boundary was designed for specific detection of transcripts lacking exon 6. Relative expression is expressed as 2−dCT, where dCT = (cycle threshold for the transcript of interest) – (cycle threshold for the house-keeping gene transcript).
