Consistent with the partitioning of heritability, a set of 2,932 genes that are highly constrained and show high intolerance to loss of function91 showed significant association with ADHD (β = 0.062, P = 2.6 × 10−4; Supplementary Table 10). We also find little evidence for effects in previously proposed candidate genes for ADHD62; of the nine proposed genes only SLC9A9 showed weak association with ADHD (P = 3.4 × 10−4; Supplementary Table 11). None of the Gene Ontology gene sets were significant after correcting for multiple testing, although the most associated included interesting nominally significant pathways such as “dopamine receptor binding” (P = 0.0010) and “Excitatory Synapse” (P = 0.0088; Supplementary Data 5).
