Reverse transcription PCR (RT-PCR) analyses for iPSC and embryoid body (EB) characterization were as described previously [30,31], using the same primers, except for PAX6, FLK1 and GATA2 which are listed in Part A of S1 Table. For all quantitative reverse transcription PCR (qRT-PCR) analyses, complementary DNA (cDNA) synthesis was performed using High Capacity cDNA reverse transcriptase kit (Life Technologies), according to the manufacturer’s instructions. Quantitative PCR analysis of reverse transcribed cDNA was conducted in triplicate using the Applied Biosystems ABI 7900 HT system with TaqMan® Assays and TaqMan® Gene Expression Master Mix (Life Technologies, Carlsbad, CA) with primer probes as listed in Part B of S1 Table.
