Human promoter sequences for APP, APLP1, and APLP2 were aligned using CLUSTAL multiple alignment algorithms and the MUSCLE (3.8) program. Nucleotides shared by all three sequences are indicated by an asterisk. The AP-1 consensus binding sequence is boxed, and the transcription start site (+1) marked in bold and boxed. The sequences targeted by sgRNAs (sg1–6) for APP CRISPRi are underlined.
