chain was found to establish a direct interaction with the LPHN3-OLF Tyr245 in the complex structure, an Ala mutant was also designed to get insights into the importance of this specific position for the affinity between the two partners. For the Phe160Ala mutant, a KD of ~1 μM was determined, suggesting that Phe160 is a key residue in the association and that the contact this residue establishes with LPHN3-OLF Tyr 245 is likely to be of major importance (Figure 5H and Table 1).
