HEK 293T and HepG2 cells (CRISPRi experiments) and NIH 3T3 cells (testing of gRNAs) were seeded into 6-well plates and transfected 24 hours later using Lipofectamine 3000 (Thermo Fisher Scientific) or TransIT-2020 Reagent (Mirus Bio) according to the manufacturer’s instructions.
