In the locus of IL-4, we identified ten regions that were differentially methylated between CPA and controls, three of these regions were found in T cells. Two of these regions that are less methylated in the T cells of CPA individuals, contain several ENCODE regulatory elements (Figure 2A bottom, Figure S2A), including a DNase hypersensitive cluster, enrichment for histone marks H3K4Me3 and H3K27Ac, and several transcription factor binding sites (Figure 2A bottom, highlighted in blue). Theses regions are located in the Th2 cytokine Locus Control Region (LCR) known to regulate the expression of IL-4 locus.
