To generate AAVs, plasmids containing cDNAs of the mouse Vkorc1, Ube2l3, Acaa2, Cpne1, and Ergic3 genes were commercially obtained (GE Dharmacon). The nucleotide sequences of the cDNA clones used in this study can be accessed using the following identifiers: Vkorc1 (BC031732), Ube2l3 (BC093503), Acaa2 (BC028901), Cpne1 (BC057554), Ergic3 (BC043720). Coding sequences were subcloned into the pENN.AAV.TBG.PI.rBG vector (Penn Vector Core), and AAV serotype 8 particles expressing the candidate genes under the control of the liver-specific TBG promoter were prepared by the Penn Vector Core at the University of Pennsylvania. Empty vectors were used as controls.
