Data from the overlapping Alzheimer’s disease datasets also overlaps significantly data from the overlapping NIDA addiction vulnerability samples (Samples 1 and 2) [12]. None of 100,000 Monte Carlo simulation trials identifies as many overlapping genes as those identified by these true datasets (Monte Carlo p < 0.00001). These comparisons thus strongly support the “transitive” genetic overlaps between addiction, cognitive function/brain volume and AD based on pleiotropic effects of variants in a number of genes.
