AMPA and NMDA receptor channel blockers CNQX and D-APV, spontaneous IPSCs were reliably detected (Fig. 4c, upper panel). Evoked IPSCs could be blocked by further addition of picrotoxin (Fig. 4c, middle panel). Similarly, at a holding potential of −70 mV and in the presence of picrotoxin, fast-decaying EPSCs mediated by AMPA-receptors could be evoced (Fig. 4d, middle panel). Conversely, at a holding potential of +60 mV (which relieves the voltage-dependent blockade of Mg2+ to NMDA-receptors), slow-decaying NMDA-receptor mediated EPSCs could be recorded (Fig. 4d, middle panel).
