We applied real-time qPCR analyses to three cohorts of mice. In the first group, we examined gene responses for three key genes from the same male mice (11 week treatment group) that were used for microarray analysis. These were compared with the 36 week females. These Cyp1b1-ko mice exhibited appreciable suppression of adiposity (Figure 1D). Figure 7B shows the relative gene expression for Scd1, PPARγ and CD36 in the 11 and 36 week groups. At 11 weeks, these results largely replicate the microarray data. At 36 weeks, the HFD-mediated suppression of Scd1 was so enhanced that Cyp1b1 deletion has no further effect. CD36 expression is relatively unchanged between 11 and 36 weeks of age, whereas the suppression of PPARγ is relieved in the female Cyp1b1-ko mice. The real-time qPCR also confirms the microarray expression results of Ppp1r3c and Ppp1r3g, mediators of glycogen synthesis (Figure 7C).
